    Chloroquine mechanism mitochondria

Three forms of autophagy are commonly described: macroautophagy, microautophagy, and chaperone-mediated autophagy (CMA). In macroautophagy, expendable cytoplasmic constituents are targeted and isolated from the rest of the cell within a double-membraned vesicle known as an autophagosome, which, in time, fuses with an available lysosome, bringing its specialty process of waste management and disposal; and eventually the contents of the vesicle (now called an autolysosome) are degraded and recycled.

Glycolysis has long been considered as the major metabolic process for energy production and anabolic growth in cancer cells. Although such a view has been instrumental for the development of powerful imaging tools that are still used in the clinics, it is now clear that mitochondria play a key role in oncogenesis. Background Chloroquine CQ is recognized as a potent adjuvant when combined with other chemotherapies to treat cancers. However, the effects of a single treatment of CQ on bladder cancer BC cells have not been investigated. Methods The growth and viability of CQ-treated BC cells were examined. Chloroquine CQ, a quinolone derivative widely used to treat and prevent malaria, has been shown to exert a potent adjuvant effect when combined with conventional glioblastoma therapy. Despite inducing lysosome destabilization and activating p53 in human glioma cells, the mechanisms underlying cell death induced by this drug are poorly understood.

In the extreme case of starvation, the breakdown of cellular components promotes cellular survival by maintaining cellular energy levels. In disease, autophagy has been seen as an adaptive response to stress, promoting survival of the cell; but in other cases it appears to promote cell death and morbidity.

Chloroquine inhibits autophagy and deteriorates the mitochondrial dysfunction and apoptosis in hypoxic rat neurons. Li P1, Hao L2, Guo YY3, Yang GL3, Mei H3, Li XH3, Zhai QX4. Chloroquine resistance is associated with a decrease in the amount of chloroquine that accumulates in the food vacuole, the site of action for chloroquine. The mechanism for this decreased accumulation is controversial. Some studies have shown that the decrease in drug accumulation is due to an increase in drug efflux. Mitochondrial quality control mechanisms have been implicated in protection against cardiac ischemia-reperfusion IR injury. Previously, cloxyquin 5-chloroquinolin-8-ol was identified via phenotypic screening as a cardioprotective compound.

Plaquenil & weight loss Lupus Forums at The Lupus Site Just wondering if anyone else had problems with loss of appetite and weight loss while on plaquenil. I had my dose increased to 400mg a day about three weeks ago, and since then I have no appetite whatsoever. Weight is falling off me at a rather alarming rate - at least half a stone a week.

ACR Actual-weight hydroxychloroquine dosing works in SLE, up. Dec 30, 2015 SAN FRANCISCO – Hydroxychloroquine Plaquenil dosing based on actual body weight – instead of ideal weight – is appropriate for patients with systemic lupus erythematosus SLE, according to a review of 686 lupus patients at Johns Hopkins University in Baltimore. Most were dosed by actual weight, 6.5 mg/kg up to a maximum of 400 mg/day.
