    Chloroquine with temodor

Listing a study does not mean it has been evaluated by the U. Treatment of GBM xenografts in vivo with chloroquine (CQ), an antimalarial agent, has been shown to reduce the hypoxic fraction and sensitizes tumors to radiation. Patients with a glioblastoma (GBM) have a poor prognosis with a median survival of 14.6 months after maximal treatment with a resection and chemoradiation.

Chloroquine CQ is widely known as an antimalarial and anti-rheumatoid drug. CQ is a weak, basic drug that can accumulate in acidic organelles, including lysosomes, resulting in increased vacuolar pH. Thus, CQ prevents the fusion of autophagosomes with lysosomes, inhibiting late stage autophagy. Temodar Warnings and Precautions Complications such as infections, bleeding, and anemia can occur in some people who use Temodar. These and other safety precautions should be reviewed to help minimize complications with this chemotherapy drug. Chloroquine CQ is commonly used in the treatment of malaria, a disease caused by infection with the parasite Plasmodium 1. There are several decades of clinical experience with the use of CQ for the treatment of various parasitic and immune-mediated disorders, although its mechanism of action is still under investigation 1, 2.

The most common EGFR mutation in GBM (EGFRv III) is present in 50-60% of patients whose tumor shows amplification of EGFR. Epidermal growth factor receptor (EGFR) amplification or mutation is regularly observed GBM and is thought to be a major contributor to radioresistance.

Chloroquine in Glioblastoma—New Horizons for an Old Drug. Anusheel Munshi, MD, DNB. Glioblastoma multiforme GBM is 1 of the most aggressive neoplasms of the central nervous sys- tem. GBMs usually follow an aggressive course, and patients have a median survival of 8 months to 10 months.1 These tumors have a diffusely infiltrating nature. Both adults and children should take one dose of chloroquine per week starting at least 1 week before. traveling to the area where malaria transmission occurs. They should take one dose per week while there, and for 4 consecutive weeks after leaving. The weekly dosage for adults is 300mg base 500mg salt. Patients in the Temodar+RT arm received concomitant Temodar 75 mg/m 2 once daily, starting the first day of RT until the last day of RT, for 42 days with a maximum of 49 days. This was followed by 6 cycles of Temodar alone 150 or 200 mg/m 2 on Days 1 to 5 of every 28-day cycle, starting 4 weeks after the end of RT.

At times I don't hear it or I'm not aware of it, we need to be rementioning this more to our rheumatologist.
