It may have both an anti-spirochaete activity and an anti-inflammatory activity, similar to the treatment of rheumatoid arthritis. And caution is required if patients have certain heart conditions, diabetes, psoriasis etc.

Chloroquine is thought to exert its antimalarial effect by preventing the polymerization of toxic heme released during proteolysis of hemoglobin in the Plasmodium digestive vacuole. The mechanism of this blockade has not been established. We incubated cultured parasites with subinhibitory doses of 3Hchloroquine and 3H quinidine. These 3Hquinoline compounds became associated with hemozoin. The mechanisms of action of hydroxychloroquine and chloroquine remain under continuous study in modern molecular medicine 17,18 using advanced tools in computational biology 19, synthetic biology. Chloroquine binds to heme or FP to form the FP-chloroquine complex; this complex is highly toxic to the cell and disrupts membrane function. Action of the toxic FP-chloroquine and FP results in cell lysis and ultimately parasite cell autodigestion. In essence, the parasite cell drowns in its own metabolic products.

The most serious adverse effects affect the eye, with dose-related retinopathy as a concern even after hydroxychloroquine use is discontinued. The most common adverse effects are a mild nausea and occasional stomach cramps with mild diarrhea.

  4. Mechanism of Action Chloroquine, a 4-aminoquinoline, is an anti-protozoal agent. The precise mechanism by which chloroquine exhibits activity is not known. Chloroquine, may exert its effect against Plasmodium species by concentrating in the acid vesicles of the parasite and by inhibiting polymerization of heme.

What is Aralen chloroquine, and how does it work mechanism of action? Chloroquine, brand name Aralen, is an anti-malarial drug. It is similar to hydroxychloroquine Plaquenil, and is useful in treating several forms of malaria as well as amebiasis that has spread outside of the intestines. Chloroquine is an aminoquinolone derivative first developed in the 1940s for the treatment of malaria. It was the drug of choice to treat malaria until the development of newer antimalarials such as pyrimethamine, artemisinin, and mefloquine. Chloroquine was granted FDA Approval on 31 October 1949. Mechanism of action Chloroquine basic concentrates in parasite food vacuole acidic. Prevent heme polymerization into hemozoin causing heme accumulation toxic to parasite. Drug complex with heme disrupt cell membrane function. Other mechanism include intercalating of parasite DNA, DNA synthesis inhibition. 7.

Plaquenil - I have been taking Plaquenil 400 mg per day for about 2 years now due to lupus flare. When i first started it, I was also started on prednisone to get my inflammation under control. Actually I started on 200 mg per day and about 2 months before tapering off pred, he upped me to 400 mg per day of plaquenil to give it plenty of time to kick in.

What is the correct way to discontinue use of plaquenil?

Contemporary treatment of systemic lupus erythematosus an update for. The prognosis for patients with systemic lupus erythematosus SLE has improved significantly, with 20-year survival now approximately 80% owing partly to effective treatment. SLE treatment has evolved from the use of conventional drugs such as hydroxychloroquine.
